Mechanism of Action:
  • Inhibits fungal cytochrome P450-dependent enzymes, leading to inhibition of ergosterol synthesis and disruption of fungal cell membrane integrity
Use:
  • Treatment of systemic fungal infections such as aspergillosis, blastomycosis, histoplasmosis, and candidiasis
  • Treatment of onychomycosis (fungal nail infection)
  • Prophylaxis of fungal infections in immunocompromised patients
Side Effects:
  • Nausea, vomiting, diarrhea
  • Headache, dizziness
  • Rash, itching
  • Liver toxicity
  • Heart failure (rare)
Contraindications:
  • Hypersensitivity to itraconazole or other azole antifungals
  • Concomitant use with certain medications such as cisapride, dofetilide, pimozide, quinidine, and ergot alkaloids
